چکیده

29 Maize was genetically engineered for the biosynthesis of the high value carotenoid 30 astaxanthin in the kernel endosperm. Introduction of a -carotene hydroxylase and a 31 carotene ketolase into a white maize genetic background extended the carotenoid pathway to 32 astaxanthin. Simultaneously, phytoene synthase, the controlling enzyme of carotenogenesis, 33 was over-expressed for enhanced carotenoid production and lycopene -cyclase was knocked34

Identification and characterization of the maize arogenate dehydrogenase gene family

In plants, the amino acids tyrosine and phenylalanine are synthesized from arogenate by arogenate dehydrogenase and arogenate dehydratase, respectively, with the relative flux to each being tightly controlled. Here the characterization of a maize opaque endosperm mutant (mto140), which also shows retarded vegetative growth, is described The opaque phenotype co-segregates with a Mutator transpos...

Central metabolic fluxes in the endosperm of developing maize seeds and their implications for metabolic engineering.

¹⁴C labeling experiments performed with kernel cultures showed that developing maize endosperm is more efficient than other non-photosynthetic tissues such as sunflower and maize embryos at converting maternally supplied substrates into biomass. To characterize the metabolic fluxes in endosperm, maize kernels were labeled to isotopic steady state using ¹³C-labeled glucose. The resultant labelin...

Cloning and Functional Characterization of the Maize (Zea mays L.) Carotenoid Epsilon Hydroxylase Gene

The assignment of functions to genes in the carotenoid biosynthesis pathway is necessary to understand how the pathway is regulated and to obtain the basic information required for metabolic engineering. Few carotenoid ε-hydroxylases have been functionally characterized in plants although this would provide insight into the hydroxylation steps in the pathway. We therefore isolated mRNA from the...

Characterization of 3-Hydroxy-3-Methylglutaryl Coenzyme A Reductase Activity during Maize Seed Development, Germination, and Seedling Emergence.

Many isoprenoid compounds are necessary for growth and development of the seed and seedling. The first committed step in the biosynthesis of isoprenoid compounds is the conversion of 3-hydroxy-3-methylglutaryl coenzyme A to mevalonate. This study shows that the specific activity of the rate-limiting enzyme, 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) is developmentally regulated duri...

Genome mining of astaxanthin biosynthetic genes from Sphingomonas sp. ATCC 55669 for heterologous overproduction in Escherichia coli

As a highly valued keto-carotenoid, astaxanthin is widely used in nutritional supplements and pharmaceuticals. Therefore, the demand for biosynthetic astaxanthin and improved efficiency of astaxanthin biosynthesis has driven the investigation of metabolic engineering of native astaxanthin producers and heterologous hosts. However, microbial resources for astaxanthin are limited. In this study, ...
